Polyclonal antibody raised to the phosphopeptide sequence YNRTRRIS(PO4)QT. Recognises Ser-2809 phosphorylated RYR2
exclusively, and will not react with dephosphorylated RYR2. Reconstitution
& Storage:
Store dessicated at 4C prior to reconstitution.
Dissolve lyophilised serum in 50ml
deionised water. Freeze in aliquots
at –70C. Avoid freeze-thaw cycles
- this denatures antibody
molecules. Best Before Date: This antibody preparation stored as described on
this sheet is very stable. It will display the characteristics described on this
sheet if used before 31 Mar 2004. Species
reactivity:
Dog, rat, sheep. [A number of customers have reported that this product will also detect
Ser-2808 phosphorylated forms of RYR2 in samples of human cardiac muscle
although evidence for this is not in Badrilla's possession.] Quality control testing
Immunoblot analysis: RYR2-PS2809 (1:5000), of this lot, detected
Ser-2809 phospho-RYR2 in 10mg
canine cardiac SR but not
dephosphorylated RYR2 in the same SR preparation (figure). All immunoreactivity
was absent when blots were probed by RYR2-PS2809 in the presence of 1mM
epitope peptide (P010-30), but addition of 1mM dephosphorylated peptide (P010-35) had no effect on
antibody binding to phosphorylated RYR2.
